I'm not sure, I went on one after Sunday mass (around noon, I think). Wonderful place, wonderful paintings.

I would say that you may rent-a-car in Seville but I strongly advise you to stay one night in Guadalupe as the round trip to Seville is more than 600 km and the monastery has fixed visit hours.
