Castello is the largest of the six sestieri (districts), and the only one not facing onto the Grand Canal. Whatever hotel you are chosing, No cars are admitted in Venice. It is possible to leave the car in the parkings around piazzale Roma or I'll Tronchetto (fee around 25/30 euros per day). Otherwise you can save some money by parking in Mestre (the fee is around 16 euros per day) and from there you can reach Venice by train (few mins, trains every 5 mins). This does not depend on the Locandiera but it is a general statement regarding the wonderful city of Venice.
