From JFK the Airtrain will take you to Sutphin Blvd/Archer Ave, where your only choice if you want to avoid changing trains, is to take the E-train (which will approach Manhattan from Queens, and enter the island from the northern side of Midtown).
The closest stop on the E-train to our hotel is 42nd Street-Port Authority, which is on 42nd Street and 8th Avenue. Our hotel is on 45th Street and 6th Avenue, so 2 avenues and 3 streets away (or a 5 to 10 minute walk depending on your suitcases).
