Driving Florence to Rome - Where to stay in Tuscany?

Hi! I'm planning a two-week honeymoon. My tentative plan is: After spending two or three days in Florence, rent a car and spend 3 days exploring Tuscany before ending up in Rome. All recommendations welcome. Which is the best town/area to stay for those three days? I would like to explore the hill towns and one or two wineries. It seems like staying at one centralized location for all three nights makes the most sense. Any specific recommendations on wineries/hotels/restaurants or itinerary would be greatly appreciated. Siena, San Gimignano, Pienza, Montepulciano, and Monticchielo are on my list, as well as the natural springs in Saturnia. Any other towns that are absolute musts or glaring oversights logistically with my tentative plan?

If you are going to Rome explore beyond Tuscany. Northern Lazio and southern Umbria are not on the tabloid travel list and thus not heavily occupied with tourists.

Wander three maps at one time: google, Booking.com and Airbnb. Venture beyond the well known!
