Hiking to Nuqui from Quibdo??

Is this a realistic possibility? google maps has a few villages labelled in between that must be connected by foot path. Obviously would be a self-sufficient multi day trek.

No, I don't think so. The Choco is still one of Colombia's poorest and most lawless provinces. A Gringo trekking through an active conflict zone may not be the best idea. Any attempt at navigating the Choco's dense jungle, would more than likely require a guide of some sort. Sure, there may be some game trails or something like that, but as an outsider, you probably won't know which ones to take. Most of the traffic in that region is via river travel and not overland. The bad things that could befall you are just too numerous to count. Drug smuggling, illegal mining, armed groups of questionable legality, not to mention possibly hostile indigenous communities

This would be one of those deals where, if anything bad happened to you, everyone would blame you almost as much as the people who did it. That’s how bad an idea it is.

There is a lot of beautiful (and safe) treks to do in Colombia (Cocuy, Los Nevados, Ciudad Perdida, Cerro Kenedy, Caño Cristales... etc). I would not advise to do this.
